WHat did you do, transfer money from your bank to pay pal? If you did, you can call the bank to stop the transfer.

Normally for me it takes a few days. Before Friday is most likely not going to happen if you started the transfer today but you may get it on Friday if your lucky. More then likely you will get your money on Saturday. I advise you get a friend to loan you money until then.
Funny how when you owe PayPal they take the money in a heartbeat but when you request to get your own funds they take their sweet time. Its not rational to leave large amounts of money in Paypal if you are ever likely to need the money ASAP. Next time just put your money in your bank account right away as it really doesn't matter as you most likely don't have a money market account with Paypal so you don't earn interest.

Transfer the money from paypal to your account.
Use a debit card to buy your sisters present. This way, the money you spend on your debit card wont come out of your account until later, and by then your money will have been transferred from PayPal.
et voilá
EDIT: Also, there shouldn't be a delay in your payment because your tax and P45 were messed up. You would only get taxed more, which you can claim back as a rebate by contacting the Inland Revenue. If it is your bosses fault, insist they sub you. They are the ones afterall, that have left you short.
