Depends on the game. If it's a Steam only game, then you can use the key to activate a game through Steam and start the download, but then exit and copy all the necessary files to the appropriate directory in the Steam folder (\Steam\steamapps\[game name]) and restart the download so it could finish things up.
If you have the legit DVD, that process is sort of automated.
Generally though, Steam does not accept retail keys, you can only install the game as retail and add it to Steam as a convenience - it won't be a Steam game, but you can just launch it from there. Sort of like a shortcut. At least you used to be able to do that, not sure about now.
Well I bought the game on steam, but I have the skidrow release on a HD, funny enough tho, the steam key worked with the game when I installed it haha.
[edit] That worked! Installed the game off the skidrow release and copied it over Now it's just updating the game

Don't know about most of today's games but the first game I bought and installed on Steam was the retail version of Half-Life 2, that came with 5 CDs but also required Steam to install and play.
Still, I think that most Steam games are usually acquired digitally nowadays. I tried linking to Steam my Guild Wars Platinum and Guild Wars Nightfall, both retail, CD-KEYs but they didn't work.
