Featuring gameplay that automatically adjusts to the player’s skill level and innovative game design that makes reading music visually fun and intuitive, Rocksmith engages experienced musicians and those who are picking up a guitar for the first time. Today, Ubisoft announced the development of Rocksmith, a new video game franchise that is set to revolutionize the music genre. This is how guitar games should be, i'm really interested. If it ends up turning out good i'll be buying it for sure. I think it'll be a blast learning new songs this way.

It doesn't seem like any hardware (other than a guitar) will be needed for this, but who knows. Here is what the official FAQ says:
Can I use my guitar with Rocksmith?
Most likely, the answer is yes. If your guitar has a standard ¼ inch jack for you to plug in a cable, then yes, your guitar will work. If your guitar does not have a jack (for example, many acoustic guitars), you can go down to your local music store and buy a pickup, and your guitar will be ready to plug in and play.
Will I need an amplifier to play the game?
Nope. All you need is a TV, a guitar, and our game. With Rocksmith, your TV or home theatre system becomes your amplifier. You plug your guitar directly into the console, and play. It's that easy.

this adapter, from audio jack to usb.
probably doesn't do much, but if they are not too stupid. it will have some kind of verification to prevent knockoffs and DIY adapters in order to decrease piracy.

You can buy audio jack to usb adapters, but it makes since what you say, they very well might force you into there adapter.

Well, well, this is interesting, I already plug my guitar into the computer as I use Guitar rig+reaper, the game looks like an uglier version of guitar hero.

I love the concept. Instead of learning how to push coloured buttons this actually gives you real transferable skills. If only they would let me play the drums.

you can play rock band and guitar hero with midi drums -> playing real drums.

We should already consider ourselves lucky that the game is about guitars, which automatically excludes pop crap a-la Gaga
