Doest it seem to fire at all when you try and turn it over?
The basics you probably know but here the first things to try.
1. Spark, are the plugs sparking when you turn it over.
2. Fuel, is the carb filling up with fuel, is it getting to much fuel and flooding? Is the fuel freely flowing from gas tank to filter(If available) to carb and onto the cylinder?
3. Air, is the air filter clean, anything obstructing air to the engine.
If you can verify all of the above, make sure its fire when you try and turn it over, if it is, it could be something as simple as air fuel mixture (Adjust the needles on the carb) or something bigger like a timing issue etc.
Im no mechanic, but i have had dirt bikes for almost my entire life. Odds are its something simple like one of the 3.

North American motorcycles have a fuel shutoff valve.
By chance have you switched off the fuel?

our cars have it too.
This may be dumb but check all your fuses. If you blow a fuel pump fuse you get no fuel.
