DX10 is a feature built into the card with backward compatibility. It'll play DX9, 8 etc games. In order to use DX10 you have to have a card (geforce 8 series and later and whatever ATI ones do) and use the windows vista OS. You'll also need a game to have DX10 programmed into it to use it as well.
You'll generally need a PCI-E slot yes since that's what all the video cards use, I believe there was a rumor though that evga I think it was, is going to make an AGP 8 series card.
